Meta Description: This document outlines the Performance Qualification Protocol for the Aqueous Buffer Preparation Tank used in NDDS for Liposomes and Lipid Nanoparticles in R&D and Production.

Tags: Performance Qualification, Equipment Validation, NDDS, Liposomes, Lipid Nanoparticles, Aqueous Buffer Preparation Tank

Objective

The objective of this Performance Qualification (PQ) is to verify that the Aqueous Buffer Preparation Tank operates within defined parameters to ensure the preparation of aqueous phase for NDDS products meets quality standards.

Scope

This protocol applies to the Aqueous Buffer Preparation Tank used in the R&D/Production areas for the preparation of aqueous phases in the manufacturing of Liposomes and Lipid Nanoparticles.

Responsibilities

The following personnel are responsible for the execution and oversight of this PQ:

  • Validation Lead: Oversee the PQ execution and ensure compliance with the protocol.
  • Quality Assurance: Review and approve the PQ documentation.
  • Production Personnel: Execute the test cases as outlined in this protocol.

Prerequisites

  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
  • Availability of necessary materials and equipment.
  • Training of personnel on the equipment operation and PQ procedures.

Equipment Description

The Aqueous Buffer Preparation Tank is designed to prepare the aqueous phase for liposomal formulations. It is equipped with temperature control, pH measurement, and conductivity monitoring capabilities.

Test Plan

Test ID Procedure Acceptance Criteria Evidence
PQ-01 Verify pH measurement accuracy pH within range 6.5 – 7.5 pH log records
PQ-02 Check conductivity levels Conductivity within range 0.5 – 1.5 mS/cm Conductivity log records
PQ-03 Monitor temperature during operation Temperature maintained at 20 – 25°C Temperature log records
PQ-04 Verify mixing records Mixing time as per SOP Mixing log records

Detailed Test Cases

Test Case PQ-01: Verify pH Measurement Accuracy

Procedure: Measure the pH of the prepared aqueous phase using a calibrated pH meter.

Acceptance Criteria: pH must be within the range of 6.5 – 7.5.

Evidence: Document pH log records and calibration certificates.

Test Case PQ-02: Check Conductivity Levels

Procedure: Measure the conductivity of the prepared aqueous phase using a calibrated conductivity meter.

Acceptance Criteria: Conductivity must be within the range of 0.5 – 1.5 mS/cm.

Evidence: Document conductivity log records and calibration certificates.

Test Case PQ-03: Monitor Temperature During Operation

Procedure: Monitor the temperature of the aqueous phase during preparation.

Acceptance Criteria: Temperature must be maintained between 20 – 25°C.

Evidence: Document temperature log records.

Test Case PQ-04: Verify Mixing Records

Procedure: Ensure that the mixing process adheres to the established SOP.

Acceptance Criteria: Mixing time must comply with the SOP.

Evidence: Document mixing log records.

Deviations

Any deviations from the acceptance criteria must be documented and investigated. A corrective action plan should be implemented as necessary.
