Performance Qualification Protocol for Depyrogenation Tunnel

This document outlines the Performance Qualification (PQ) protocol for the Depyrogenation Tunnel used in the production of prefilled syringes and cartridges.

Objective: To validate that the Depyrogenation Tunnel effectively reduces endotoxin levels in syringes and cartridges to meet specified acceptance criteria.

Scope: This protocol applies to the Depyrogenation Tunnel utilized in the production area for the depyrogenation of prefilled syringes and cartridges.

Responsibilities:

  • Validation Team: Execute the PQ protocol and document results.
  • Quality Assurance: Review and approve the PQ results and protocol.
  • Production Team: Ensure proper operation of the Depyrogenation Tunnel during testing.

Prerequisites:

  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
  • Training of personnel on the operation of the Depyrogenation Tunnel.
  • Availability of all necessary equipment and materials for testing.

Equipment Description: The Depyrogenation Tunnel is designed to effectively eliminate endotoxins from prefilled syringes and cartridges through controlled heat treatment. Key parameters include temperature profile, belt speed, and PLC logs.

Test ID Procedure Acceptance Evidence
PQ-01 Verify temperature profile throughout the tunnel. Temperature within specified limits. Temperature logs.
PQ-02 Measure belt speed during operation. Belt speed within specified limits. Belt speed logs.
PQ-03 Conduct endotoxin testing on samples. Endotoxin levels below specified threshold. Endotoxin test results.
PQ-04 Review PLC logs for consistency. Logs show no anomalies. PLC log reports.

Detailed Test Cases:

  • Test Case PQ-01: Monitor the temperature at multiple points in the tunnel to ensure the profile remains within the validated range during operation.
  • Test Case PQ-02: Verify the belt speed using calibrated measuring devices and ensure it is consistent with the validated speed.
  • Test Case PQ-03: Collect samples post-depyrogenation and conduct endotoxin testing using the Limulus Amebocyte Lysate (LAL) method.
  • Test Case PQ-04: Review PLC logs for any discrepancies or errors that may indicate a malfunction during the process.

Deviations: Any deviations from the protocol must be documented and investigated. A root cause analysis should be performed, and corrective actions should be implemented as necessary.

Data Integrity Checks:

  • Ensure all logs are timestamped and signed by the operator.
  • Regularly back up PLC logs to prevent data loss.
  • Conduct periodic audits of data entries for accuracy.
