Purpose

The purpose of this SOP is to establish a standardized approach for the validation of the High Shear Mixer/Homogenizer used in the production of Transdermal Patches, ensuring uniform drug distribution in the adhesive matrix.

Scope

This SOP applies to the validation of the High Shear Mixer/Homogenizer in the Production area for the manufacturing of Transdermal Patches.

Definitions

  • High Shear Mixer/Homogenizer: Equipment used to achieve uniform dispersion of active pharmaceutical ingredients in the adhesive matrix.
  • Validation: Documented evidence that a system performs its intended function consistently and reliably.
  • CSV: Computerized System Validation.

Roles

  • Validation Team: Responsible for planning, executing, and documenting the validation process.
  • Quality Assurance: Ensures compliance with regulatory requirements and reviews validation documentation.
  • Production Personnel: Operate the equipment and provide input during the validation process.

Lifecycle Procedure

  1. Design Qualification (DQ): Verify that the equipment design meets user requirements.
  2. Installation Qualification (IQ): Confirm that the equipment is installed correctly and according to specifications.
  3. Operational Qualification (OQ): Demonstrate that the equipment operates as intended across all specified operating ranges.
  4. Performance Qualification (PQ): Validate the equipment’s performance in a production environment.

Good Documentation Practices (GDP) Controls

All validation documents must be completed in accordance with GDP to ensure accuracy and traceability. All entries must be dated, signed, and legible.

Acceptance Criteria Governance

Acceptance criteria will be established based on User Requirements Specification (URS) Annex 11 and must be met for each qualification phase.

Calibration/PM Governance

The High Shear Mixer/Homogenizer must undergo regular calibration and preventive maintenance as per the manufacturer’s recommendations and internal procedures.

Change Control Triggers

Any changes to equipment, process, or materials that may impact the performance or validation status of the High Shear Mixer/Homogenizer must be evaluated through a change control process.

Revalidation Triggers and Periodic Review

Revalidation is required every 12 months or when significant changes occur that may affect equipment performance or product quality.
