Performance Qualification Protocol for Inline Static Mixer in NDDS Production
Meta Description: This document outlines the Performance Qualification protocol for the Inline Static Mixer used in the production of NDDS, focusing on liposomes and lipid nanoparticles.
Tags: Performance Qualification, Equipment Validation, NDDS, Liposomes, Lipid Nanoparticles, Inline Static Mixer
Objective
The objective of this Performance Qualification (PQ) protocol is to ensure that the Inline Static Mixer operates consistently and reliably within the defined parameters during the mixing process for the formation of liposomes and lipid nanoparticles.
Scope
This protocol applies to the Inline Static Mixer used in the R&D and Production areas for the mixing of liposomes and lipid nanoparticles. The focus is on the critical parameters affecting product quality.
Responsibilities
- Validation Team: Responsible for executing the PQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the PQ protocol and results.
- Production Team: Responsible for operating the Inline Static Mixer during the qualification process.
Prerequisites
- Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
- Availability of Standard Operating Procedures (SOPs) for the Inline Static Mixer.
- Training of personnel on the operation of the Inline Static Mixer.
Equipment Description
The Inline Static Mixer is designed for the efficient mixing of materials during the formation of liposomes and lipid nanoparticles. It operates without moving parts, utilizing a series of static mixing elements to achieve homogeneous mixing.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| PQ-001 | Measure pressure drop during mixing at specified flow rates. | Pressure drop within URS specifications. | Pressure drop data logs. |
Detailed Test Cases
Test Case PQ-001
Objective: To evaluate the mixing efficiency by measuring the pressure drop across the Inline Static Mixer.
Procedure:
1. Set up the Inline Static Mixer according to the manufacturer’s instructions.
2. Begin the mixing process at the defined flow rates.
3. Record the pressure drop across the mixer using calibrated pressure gauges.
4. Repeat the measurement for three consecutive runs to ensure consistency.
Acceptance Criteria: The pressure drop must remain within the specifications outlined in the User Requirement Specification (URS).
Evidence: Documented pressure drop readings and graphs for analysis.
Deviations
Any deviations from the established protocol must be documented, including the reason for deviation, impact assessment, and corrective actions taken.
Approvals
This protocol requires approval from the Quality Assurance department and must be signed by the respective approvers.
