Standard Operating Procedure for the Validation of Patch Coating Machine in Transdermal Patch Production
Purpose: This SOP outlines the validation process for the Patch Coating Machine used to apply the drug adhesive layer onto the backing film in the production of transdermal patches, ensuring compliance with regulatory standards and product quality.
Scope: This procedure applies to the validation of the Patch Coating Machine (Knife-over-roll / Slot Die) utilized in the production area for transdermal patches. It includes all phases of validation: Design Qualification (DQ), Installation Qualification (IQ), Operational Qualification (OQ), and Performance Qualification (PQ).
Definitions:
- Validation: The process of demonstrating that a system meets its intended use and produces consistent results.
- Patch Coating Machine: Equipment used to coat the drug adhesive layer onto the backing film.
- Criticality: The importance of the equipment in ensuring product quality; in this case, it is classified as Critical.
- CSV: Computer System Validation, necessary for systems that control or monitor the manufacturing process.
Roles:
- Validation Team: Responsible for planning and executing the validation activities.
- Quality Assurance (QA): Ensures compliance with regulatory requirements and oversees the validation process.
- Production Personnel: Operate the equipment and provide feedback during the validation process.
Lifecycle Procedure:
- Design Qualification (DQ): Document requirements and specifications for the Patch Coating Machine.
- Installation Qualification (IQ): Verify that the equipment is installed according to specifications.
- Operational Qualification (OQ): Test the equipment under operational conditions to ensure it performs as intended.
- Performance Qualification (PQ): Validate that the equipment consistently produces the desired output.
GDP Controls: Good Documentation Practices must be followed throughout the validation process to ensure traceability and compliance.
Acceptance Criteria Governance: Acceptance criteria will be governed by User Requirement Specifications (URS) and must comply with Annex 11 and Annex 15 of the relevant guidelines.
Calibration/PM Governance: The Patch Coating Machine must undergo regular calibration and preventive maintenance as per the manufacturer’s recommendations and internal procedures.
Change Control Triggers: Any modifications to the equipment, process, or materials that may impact the validation status must be documented and assessed through the change control process.
Revalidation Triggers and Periodic Review: Revalidation is required every 12 months or whenever a significant change occurs in the process, equipment, or materials.
