Performance Qualification Protocol for Liposome Extruder
Meta Description: This document outlines the Performance Qualification (PQ) protocol for the Liposome Extruder used in the R&D and production of Liposomes and Lipid Nanoparticles.
Tags: Performance Qualification, Equipment Validation, Liposomes, Lipid Nanoparticles, Extrusion System
Objective
The objective of this Performance Qualification (PQ) protocol is to ensure that the Extrusion System operates consistently and reliably within specified limits for the production of Liposomes and Lipid Nanoparticles.
Scope
This protocol applies to the Liposome Extruder used in the R&D and production areas for size control via membrane extrusion. It covers the validation of critical parameters that impact product quality.
Responsibilities
- Validation Team: Responsible for executing the PQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the PQ protocol and results.
- Equipment Operator: Responsible for operating the Extrusion System as per the protocol.
Prerequisites
- Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
- Training of personnel on the Extrusion System operation.
- Availability of all necessary materials and equipment for testing.
Equipment Description
The Liposome Extruder is designed for the production of liposomes and lipid nanoparticles through membrane extrusion technology. It operates under controlled pressure and temperature conditions to ensure consistent product quality.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| TP-01 | Measure pressure during extrusion | Pressure within specified range | Pressure log records |
| TP-02 | Check membrane pore size | Pore size meets specifications | Membrane specifications report |
| TP-03 | Monitor temperature during operation | Temperature within specified limits | Temperature log records |
| TP-04 | Measure flow rate | Flow rate meets specifications | Flow rate records |
Detailed Test Cases
Test Case 1: Pressure Measurement
Objective: To verify that the extrusion pressure is maintained within specified limits.
Procedure: Record pressure readings at regular intervals during the extrusion process.
Acceptance Criteria: Pressure must remain within the specified range as defined in the URS Annex11.
Test Case 2: Membrane Pore Size Verification
Objective: To confirm that the membrane used in the extrusion system meets pore size specifications.
Procedure: Use a calibrated measurement tool to assess the pore size of the membrane.
Acceptance Criteria: Pore size must match the specifications provided in the URS Annex11.
Test Case 3: Temperature Monitoring
Objective: To ensure that the temperature during operation is controlled and within acceptable limits.
Procedure: Continuously monitor and log the temperature during the extrusion process.
Acceptance Criteria: Temperature readings must remain within the defined limits as per URS Annex11.
Test Case 4: Flow Rate Measurement
Objective: To verify that the flow rate of the material through the system is consistent with specifications.
Procedure: Measure and record the flow rate during the extrusion process.
Acceptance Criteria: Flow rate must adhere to the specifications outlined in the URS Annex11.
Deviations
Any deviations from the acceptance criteria must be documented and reviewed. Corrective actions should be initiated as necessary to address any issues identified during the testing process.
Approvals
This PQ protocol must be reviewed and approved by the Quality Assurance team prior to execution. All results must be signed off by the validation team and Quality Assurance upon completion of the protocol.
