Objective
The objective of this Installation Qualification (IQ) Protocol is to verify that the Tangential Flow Filtration (TFF) System is installed according to manufacturer specifications and is suitable for use in R&D and production for buffer exchange and concentration of Liposomes and Lipid Nanoparticles.
Scope
This protocol applies to the Tangential Flow Filtration (TFF) System located in the R&D/Production area, used for direct impact on product quality.
Responsibilities
The Validation Team is responsible for executing this protocol, while the Quality Assurance (QA) team will oversee the process and ensure compliance with regulatory standards.
Prerequisites
1. Equipment must be delivered and unpacked.
2. Installation site must meet environmental and utility requirements.
3. Personnel must be trained on equipment operation and safety procedures.
Equipment Description
The Tangential Flow Filtration (TFF) System is designed for buffer exchange and concentration of Liposomes and Lipid Nanoparticles. It features a membrane filtration system capable of maintaining critical parameters such as TMP, flow rate, and membrane integrity.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| IQ-TFF-01 | Verify installation location and environmental conditions | Location meets specifications outlined in URS | Photographic evidence, checklist |
| IQ-TFF-02 | Check utility connections (water, electricity, compressed air) | All utilities operational and within required parameters | Utility test reports |
| IQ-TFF-03 | Perform functional checks on system | System operates without errors, parameters within limits | Functional test report |
| IQ-TFF-04 | Verify data integrity checks | Audit trail and data logs are complete and accurate | CSV report |
Detailed Test Cases
Test Case 1: Installation Verification
Verify that the installation location meets the specified environmental conditions, including temperature and humidity.
Test Case 2: Utility Connection Check
Confirm that all necessary utilities are connected and functioning properly. Document any discrepancies.
Test Case 3: Functional Testing
Run the TFF system and monitor critical parameters such as TMP and flow rate. Document results against acceptance criteria.
Test Case 4: Data Integrity Checks
Ensure that all data logs are generated correctly, and the audit trail is intact. Perform checks to verify data accuracy.
Deviations
Any deviations from the acceptance criteria must be documented, investigated, and resolved prior to equipment qualification.
Approvals
All test results must be reviewed and approved by the Validation Team and Quality Assurance prior to proceeding with the next phases of validation.
