Performance Qualification Protocol for Depyrogenation Tunnel / Dry Heat Sterilizer
Meta Description: This document outlines the Performance Qualification (PQ) protocol for the Depyrogenation Tunnel / Dry Heat Sterilizer used in the production of sterile eye drops and ointments.
Objective
The objective of this Performance Qualification (PQ) protocol is to validate the Depyrogenation Tunnel / Dry Heat Sterilizer for the sterilization and depyrogenation of glass bottles used in the production of sterile eye drops and ointments.
Scope
This protocol applies to the Depyrogenation Tunnel / Dry Heat Sterilizer located in the Production area, specifically for the sterilization of glass containers intended for direct product contact.
Responsibilities
- Validation Team: Responsible for executing the PQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the protocol and results.
- Production Personnel: Responsible for operating the equipment as per the validated procedures.
Prerequisites
- Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
- Availability of all required materials and resources.
- Training of personnel on equipment operation and safety procedures.
Equipment Description
The Depyrogenation Tunnel / Dry Heat Sterilizer is designed to sterilize and depyrogenate glass bottles through controlled temperature profiles, ensuring effective endotoxin reduction.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| TP-01 | Temperature profile mapping | Temperature to be maintained as per URS Annex 15 | Temperature logs |
| TP-02 | Belt speed verification | Belt speed as per established parameters | Belt speed measurement |
| TP-03 | Dwell time assessment | Dwell time meets defined criteria | Dwell time logs |
| TP-04 | Endotoxin reduction testing | Endotoxin levels reduced as per specifications | Endotoxin test results |
Detailed Test Cases
Test Case 1: Temperature Profile Mapping
Conduct a temperature mapping study to ensure uniform temperature distribution throughout the tunnel. Utilize data loggers placed at critical points.
Test Case 2: Belt Speed Verification
Measure the belt speed using a calibrated tachometer to ensure it operates within specified limits.
Test Case 3: Dwell Time Assessment
Verify the dwell time of containers within the sterilizer, ensuring it meets the defined specifications.
Test Case 4: Endotoxin Reduction Testing
Perform endotoxin testing on samples post-sterilization to confirm compliance with established limits.
Deviations
Any deviations from the acceptance criteria must be documented and investigated. A root cause analysis should be conducted, and corrective actions should be implemented as necessary.
Approvals
This protocol must be reviewed and approved by the Quality Assurance department prior to execution. All results must be signed off by the Validation Team and Quality Assurance.
Data Integrity Checks
- Ensure data loggers are calibrated and functioning prior to use.
- Implement electronic data capture systems with audit trails for temperature and speed readings.
- Perform routine checks and maintenance on measuring instruments to ensure accuracy.
