Design Qualification Protocol for Patch Leak Test Machine

Meta Description: This document outlines the Design Qualification Protocol for the Patch Leak Test Machine used in the validation of transdermal patches.

Tags: Equipment Validation, Design Qualification, Transdermal Patches, Patch Leak Test Machine

Objective

The objective of this protocol is to establish the requirements for the Design Qualification of the Patch Leak Test Machine, ensuring it meets specified operational and regulatory standards for verifying pouch seal integrity in transdermal patches.

Scope

This protocol applies to the Patch Leak Test Machine utilized in the Production/QC area for the verification of pouch seal integrity in transdermal patches. It encompasses all activities related to the qualification of the equipment.

Responsibilities

  • Validation Team: Responsible for executing the qualification protocol and documenting results.
  • Quality Assurance: Responsible for reviewing and approving the protocol and results.
  • Production/QC Personnel: Responsible for operating the equipment and following standard operating procedures.

Prerequisites

  • Installation of the Patch Leak Test Machine in the designated area.
  • Completion of equipment installation qualification (IQ).
  • Availability of all necessary documentation, including user requirements specifications (URS).

Equipment Description

The Patch Leak Test Machine is an instrument designed to verify the integrity of pouch seals in transdermal patches by measuring leak rates. It is equipped with features that allow for sensitive detection of leaks, ensuring product quality and compliance.

Test Plan

Test ID Procedure Acceptance Criteria Evidence
T1 Conduct leak rate sensitivity audit trail logs. Leak rate must not exceed the limits defined in URS Annex11. Audit trail log report.
T2 Perform functional tests on the machine. All functions must operate within defined parameters. Functional test report.

Detailed Test Cases

Test Case T1: Leak Rate Sensitivity Audit Trail Logs

Procedure: Run the machine under standard operating conditions and record the leak rate. Ensure that the sensitivity settings are configured correctly.

Acceptance Criteria: The leak rate must be below the maximum specified in URS Annex11.

Evidence: Printout of the audit trail log and review of the data integrity checks.

Test Case T2: Functional Tests

Procedure: Execute all machine functions as per the operational manual and verify the output against expected results.

Acceptance Criteria: All functions must operate correctly without errors.

Evidence: Functional test report and any corrective actions taken if necessary.

Deviations

Any deviations from the acceptance criteria must be documented, including the nature of the deviation, investigation, and corrective actions taken. Approval from Quality Assurance is required for all deviations.

Data Integrity Checks

As CSV is required, ensure that the following data integrity checks are implemented:

  • Validation of data entry points to prevent erroneous data input.
  • Regular audits of data logs to ensure compliance with regulatory standards.
  • Backup procedures for all data collected during testing.
