Objective
The objective of this Performance Qualification (PQ) protocol is to establish that the Tube Sealing Machine operates consistently and reliably within specified parameters to ensure the integrity of sterile eye drops and eye ointments.
Scope
This protocol applies to the Tube Sealing Machine utilized in the production and packaging areas for the sealing of tubes containing sterile ophthalmic products.
Responsibilities
The following personnel are responsible for the execution and oversight of this PQ protocol:
- Validation Team: Conducts the qualification activities.
- Quality Assurance: Reviews and approves the protocol and results.
- Production Personnel: Operate the equipment during testing.
Prerequisites
Prior to execution of this PQ protocol, the following must be completed:
- Installation Qualification (IQ) must be successfully completed.
- Operational Qualification (OQ) must be successfully completed.
- Training of personnel on the operation of the Tube Sealing Machine.
Equipment Description
The Tube Sealing Machine (Hot Air/Ultrasonic) is designed to seal tubes containing sterile ophthalmic products. Critical parameters include sealing temperature, pressure, dwell time, and the ability to generate a reject audit trail.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| PQ-001 | Verify sealing temperature | Within specified range as per URS Annex 11 | Temperature log |
| PQ-002 | Verify sealing pressure | Within specified range as per URS Annex 11 | Pressure log |
| PQ-003 | Verify dwell time | Within specified range as per URS Annex 11 | Dwell time log |
| PQ-004 | Audit trail verification | Complete and accurate audit trail generated | Audit trail report |
Detailed Test Cases
Test Case: PQ-001 – Verify Sealing Temperature
Procedure: Measure the sealing temperature using calibrated thermocouples during the sealing process.
Acceptance Criteria: The sealing temperature must be within the specified range outlined in URS Annex 11.
Evidence: Documented temperature logs must be reviewed and signed by the validation team.
Test Case: PQ-002 – Verify Sealing Pressure
Procedure: Measure the sealing pressure using calibrated pressure gauges during the sealing process.
Acceptance Criteria: The sealing pressure must be within the specified range outlined in URS Annex 11.
Evidence: Documented pressure logs must be reviewed and signed by the validation team.
Test Case: PQ-003 – Verify Dwell Time
Procedure: Measure the dwell time during the sealing process using timers.
Acceptance Criteria: The dwell time must be within the specified range outlined in URS Annex 11.
Evidence: Documented dwell time logs must be reviewed and signed by the validation team.
Test Case: PQ-004 – Audit Trail Verification
Procedure: Review the audit trail generated by the Tube Sealing Machine to ensure accuracy and completeness.
Acceptance Criteria: The audit trail must be complete and accurate.
Evidence: Audit trail report must be reviewed and signed by the Quality Assurance team.
Deviations
Any deviations from the established acceptance criteria must be documented, investigated, and approved by Quality Assurance before proceeding with the PQ protocol.
Approvals
All results from the PQ protocol must be reviewed and approved by the Quality Assurance team prior to the release of the Tube Sealing Machine for use in production.
