Installation Qualification Protocol for Spray Dryer in NDDS

Objective: To establish and document the Installation Qualification (IQ) of the Spray Dryer utilized for converting dispersion to dry powder in the production of polymeric nanoparticles.

Scope: This protocol covers the installation qualification of the Spray Dryer used in R&D and production areas for the development of NDDS – Polymeric Nanoparticles (PLGA etc.).

Responsibilities:

  • Validation Team: Ensure all validation activities are conducted in compliance with applicable regulations.
  • Equipment Owner: Provide necessary documentation and access to the equipment.
  • Quality Assurance: Review and approve the IQ protocol and results.

Prerequisites:

  • Completion of User Requirement Specification (URS).
  • Installation of Spray Dryer in the designated area.
  • Availability of necessary tools and materials for testing.

Equipment Description: The Spray Dryer is designed for the efficient conversion of dispersion into dry powder, specifically for the production of polymeric nanoparticles. It features critical parameters including inlet/outlet temperature, atomization flow, cyclone efficiency, and audit trail capabilities.

Test ID Procedure Acceptance Evidence
IQ-01 Verify installation location and environmental conditions. Location meets specifications outlined in URS. Photographic evidence and environmental log.
IQ-02 Check for completeness of equipment installation. All components installed as per manufacturer’s specifications. Installation checklist signed off by technician.
IQ-03 Validate critical parameter settings (inlet/outlet temp, atomization flow, cyclone efficiency). Settings fall within defined operational ranges. Calibration records and parameter logs.
IQ-04 Review audit trail functionality. Audit trail captures all critical operations. Audit trail report generated from the system.

Detailed Test Cases:

  • Test Case for IQ-01: Measure temperature and humidity in the installation area. Acceptable range: 20-25°C and 40-60% RH.
  • Test Case for IQ-02: Confirm that all components (pumps, nozzles, etc.) are installed according to the installation manual.
  • Test Case for IQ-03: Set inlet temperature to 150°C, outlet temperature to 50°C, atomization flow to 0.5 L/min, and verify cyclone efficiency.
  • Test Case for IQ-04: Generate an audit trail report after a test run to ensure all actions are logged.

Deviations: Any deviations from the acceptance criteria must be documented and reviewed by the validation team and Quality Assurance.
