Design Qualification Protocol for Laminar Air Flow Equipment
Meta Description: Comprehensive Design Qualification Protocol for Laminar Air Flow Equipment used in Biologics Upstream Processing, ensuring compliance and critical parameter evaluation.
Objective
The objective of this Design Qualification (DQ) protocol is to verify that the Laminar Air Flow (LAF) equipment meets the defined requirements for aseptic protection in the production/cell culture area.
Scope
This protocol applies to the Laminar Air Flow equipment used in the production and cell culture processes within the biologics manufacturing facility.
Responsibilities
Validation Team: Responsible for executing the DQ protocol and documenting results.
Quality Assurance: Responsible for reviewing and approving the DQ protocol and results.
Production Team: Responsible for providing necessary operational insights and supporting the validation process.
Prerequisites
All personnel involved in the execution of this protocol must be trained in equipment operation and validation processes. The LAF unit must be installed and operational prior to testing.
Equipment Description
The Laminar Air Flow (LAF) equipment is designed to provide aseptic protection during the handling of biological materials in a controlled environment. The critical parameters for validation include air velocity, HEPA filter integrity, and smoke study results.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| DQ-LAF-01 | Measure air velocity at various locations. | Air velocity must be within specified range as per URS. | Air velocity measurement report. |
| DQ-LAF-02 | Perform HEPA integrity test. | Integrity test must indicate no leaks. | HEPA integrity test report. |
| DQ-LAF-03 | Conduct smoke study. | No turbulence or contamination must be observed. | Smoke study video and report. |
Detailed Test Cases
Test Case 1: Air Velocity Measurement
Procedure: Measure the air velocity at 6 defined points within the LAF unit using an anemometer.
Acceptance Criteria: Air velocity must be between 0.45 m/s and 0.75 m/s.
Test Case 2: HEPA Integrity Test
Procedure: Perform a DOP test to assess the integrity of the HEPA filter.
Acceptance Criteria: No leaks should be detected during the test.
Test Case 3: Smoke Study
Procedure: Introduce smoke into the airflow and observe for turbulence.
Acceptance Criteria: No turbulence or backflow should be observed, ensuring aseptic conditions.
Deviations
Any deviations from the acceptance criteria must be documented, along with the rationale and corrective actions taken. A re-test may be required to confirm compliance.
Approvals
This protocol must be signed and approved by the Validation Team Lead and Quality Assurance prior to execution.
