Pharmaceutical

Purpose

The purpose of this SOP is to outline the validation process for the Water for Injection (WFI) System to ensure compliance with regulatory requirements and to maintain product quality.

Scope

This SOP applies to the validation of the Water for Injection System utilized in the Utilities area for pharmaceutical manufacturing. It covers all phases of validation including Design Qualification (DQ), Installation Qualification (IQ), Operational Qualification (OQ), and Performance Qualification (PQ).

Definitions

  • WFI: Water for Injection, a critical utility used in pharmaceutical manufacturing.
  • Validation: The process of establishing documented evidence that a system operates consistently and reliably.
  • DQ: Design Qualification.
  • IQ: Installation Qualification.
  • OQ: Operational Qualification.
  • PQ: Performance Qualification.

Roles

The following roles are involved in the validation process:

  • Validation Manager: Oversees the validation process and ensures compliance.
  • Quality Assurance: Reviews and approves validation documentation.
  • Engineering: Responsible for the installation and maintenance of the WFI System.
  • Operations: Conducts the operational aspects of the qualification process.

Lifecycle Procedure

  1. Design Qualification (DQ): Document design specifications and user requirements.
  2. Installation Qualification (IQ): Verify that the system is installed according to specifications.
  3. Operational Qualification (OQ): Test the system under normal operating conditions.
  4. Performance Qualification (PQ): Validate the system’s performance in producing WFI.

Good Documentation Practices (GDP) Controls

All validation documents must be completed in accordance with Good Documentation Practices. This includes clear, legible entries, proper corrections, and timely completion of records.

Acceptance Criteria Governance

Acceptance criteria for validation will be established based on User Requirements Specification (URS) and will comply with Annex 11 and Annex 15 of the EU GMP guidelines.

Calibration and Preventive Maintenance Governance

All equipment used in the WFI System must be calibrated and maintained according to the manufacturer’s specifications and internal policies to ensure consistent performance.

Change Control Triggers

Any changes to the WFI System, including modifications to equipment, processes, or materials, must initiate a change control process to assess the impact on validation status.

Revalidation Triggers and Periodic Review

The WFI System will undergo revalidation every 12 months or whenever significant changes occur. A periodic review will be conducted to ensure ongoing compliance with validation requirements.
