Terminal HEPA Filters – Equipment Validation SOP

Standard Operating Procedure for Terminal HEPA Filter Validation

Purpose

This SOP outlines the procedures for the validation of Terminal HEPA Filters used in sterile (injectables/aseptic) environments to ensure compliance with regulatory standards and product safety.

Scope

This procedure applies to the validation of Terminal HEPA Filters utilized in Grade A/B areas within the engineering department.

Definitions

  • HEPA Filter: High-Efficiency Particulate Air filter designed to remove at least 99.97% of airborne particles.
  • Grade A/B: Cleanroom classifications as per ISO 14644.
  • Validation: Establishing documented evidence that a process, system, or equipment consistently produces results meeting predetermined specifications.

Roles

  • Validation Team: Responsible for executing and documenting the validation process.
  • Quality Assurance: Ensures compliance with regulatory requirements and reviews validation documentation.
  • Engineering: Maintains the equipment and ensures proper functioning of the HEPA filters.

Lifecycle Procedure

  1. Design Qualification (DQ): Verification that the design meets user needs and intended uses.
  2. Installation Qualification (IQ): Confirmation that the equipment is installed correctly and according to specifications.
  3. Operational Qualification (OQ): Testing to ensure the equipment operates within specified limits under normal operating conditions.
  4. Performance Qualification (PQ): Verification that the equipment consistently performs as intended in a controlled environment.

Good Documentation Practices (GDP) Controls

All validation activities must be documented in accordance with GDP to ensure traceability, accuracy, and compliance.

Acceptance Criteria Governance

Acceptance criteria for HEPA filters shall adhere to the standards outlined in ISO 14644 Annex 1, ensuring that particulate levels are within acceptable limits for Grade A/B environments.

Calibration/PM Governance

Regular calibration and preventive maintenance must be scheduled every 6 to 12 months to ensure optimal performance and compliance with regulatory standards.

Change Control Triggers

Any modifications to the HEPA filter system, including replacement or significant adjustments, will initiate a change control process to evaluate potential impacts on validation status.

Revalidation Triggers and Periodic Review

Revalidation will be triggered by changes in equipment, processes, or if performance falls below established acceptance criteria. A periodic review will occur every 6 to 12 months.
