Standard Operating Procedure for Validation of pH Meter in QC Laboratory
Purpose
The purpose of this SOP is to outline the procedures for the validation of pH meters used in the QC laboratory to ensure accurate and reliable pH measurements that directly impact product quality.
Scope
This SOP applies to all pH meters used in the QC laboratory for pH measurement in pharmaceutical products. It covers the validation lifecycle including DQ, IQ, OQ, and PQ, as well as ongoing maintenance and revalidation processes.
Definitions
- DQ: Design Qualification
- IQ: Installation Qualification
- OQ: Operational Qualification
- PQ: Performance Qualification
- CSV: Computerized System Validation
- URS: User Requirement Specification
Roles
- Validation Team: Responsible for planning and executing validation activities.
- Quality Assurance: Ensures compliance with regulatory requirements and reviews validation documentation.
- Lab Personnel: Operate the pH meter and ensure proper maintenance and calibration.
Lifecycle Procedure
- Design Qualification (DQ): Document the user requirements and specifications for the pH meter.
- Installation Qualification (IQ): Verify the installation of the pH meter according to manufacturer specifications.
- Operational Qualification (OQ): Test the pH meter’s functionality to ensure it operates within specified limits.
- Performance Qualification (PQ): Confirm that the pH meter performs as intended in the actual operating environment.
Good Documentation Practices (GDP) Controls
All validation activities must be documented in accordance with Good Documentation Practices (GDP) to ensure traceability and compliance. Records must be maintained and readily accessible for review.
Acceptance Criteria Governance
Acceptance criteria for validation will be defined based on the User Requirement Specification (URS) and must comply with Annex 11 of the relevant regulations. All criteria must be met for successful validation.
Calibration and Preventive Maintenance (PM) Governance
The pH meter must be calibrated at specified intervals and maintained according to the manufacturer’s recommendations. Records of calibration and maintenance must be retained as part of the validation documentation.
Change Control Triggers
Any changes to the pH meter, its operating environment, or related processes must be assessed for impact on validation status. A change control process must be initiated for any significant modifications.
Revalidation Triggers and Periodic Review
Revalidation of the pH meter is required every 12 months or whenever a significant change occurs that may affect its performance. A periodic review of the validation status must also be conducted annually.
