Operational Qualification Protocol for Vacuum Emulsifier in Semi-Solid Production

Meta Description: This document outlines the Operational Qualification Protocol for the Vacuum Emulsifier used in the emulsification and mixing of creams, ointments, gels, and lotions in a production environment.

Objective

The objective of this protocol is to verify that the Vacuum Emulsifier operates within predefined parameters and meets the acceptance criteria as outlined in the User Requirement Specification (URS) Annex 11.

Scope

This protocol applies to the Operational Qualification of the Vacuum Emulsifier used in the production of semi-solid dosage forms including creams, ointments, gels, and lotions.

Responsibilities

The Validation Team is responsible for executing this protocol and documenting results. The Quality Assurance Team will review and approve the protocol and results.

Prerequisites

All equipment must be installed, calibrated, and maintained according to standard operating procedures (SOPs). Operators must be trained on the equipment and this protocol.

Equipment Description

The Vacuum Emulsifier is a main manufacturing vessel designed for the emulsification and mixing of creams, ointments, gels, and lotions. It operates under controlled conditions of agitation speed, temperature, vacuum, and homogenization.

Test ID Procedure Acceptance Criteria Evidence
OQ-VE-001-01 Verify Agitation Speed Agitation speed must be within the range specified in URS Annex 11. PLC logs, Calibration records
OQ-VE-001-02 Verify Temperature Control Temperature must be maintained within the specified range in URS Annex 11. PLC logs, Calibration records
OQ-VE-001-03 Verify Vacuum Levels Vacuum levels must meet specifications outlined in URS Annex 11. PLC logs, Calibration records
OQ-VE-001-04 Verify Homogenization Process Homogenization must achieve the desired particle size and distribution as per URS Annex 11. PLC logs, Test results

Detailed Test Cases

Test Case OQ-VE-001-01: Verify Agitation Speed

Procedure: Set the emulsifier to the specified agitation speed and monitor using PLC logs.

Acceptance Criteria: Agitation speed must be within the specified range in URS Annex 11.

Evidence: Documented PLC logs showing speed readings.

Test Case OQ-VE-001-02: Verify Temperature Control

Procedure: Set the emulsifier to the desired temperature and monitor using PLC logs.

Acceptance Criteria: Temperature must be maintained within the specified range in URS Annex 11.

Evidence: Documented PLC logs showing temperature readings.

Test Case OQ-VE-001-03: Verify Vacuum Levels

Procedure: Set the emulsifier to the specified vacuum level and monitor using PLC logs.

Acceptance Criteria: Vacuum levels must meet specifications outlined in URS Annex 11.

Evidence: Documented PLC logs showing vacuum readings.

Test Case OQ-VE-001-04: Verify Homogenization Process

Procedure: Conduct the homogenization process and analyze the particle size distribution.

Acceptance Criteria: Homogenization must achieve the desired particle size and distribution as per URS Annex 11.

Evidence: Particle size analysis report.

Deviations

Any deviations from the acceptance criteria must be documented and investigated. A root cause analysis will be performed for any critical failures.
