Standard Operating Procedure for Equipment Validation of Jet Mill / Micronizer

Purpose

The purpose of this SOP is to outline the validation process for the Jet Mill / Micronizer used in the production of solid dosage forms, ensuring compliance with regulatory standards and product quality requirements.

Scope

This SOP applies to the validation of Jet Mill / Micronizer equipment utilized in the production area for micronization processes. It encompasses all phases of validation including Design Qualification (DQ), Installation Qualification (IQ), Operational Qualification (OQ), and Performance Qualification (PQ).

Definitions

  • Jet Mill / Micronizer: A device used for micronization, utilizing high-velocity air jets to reduce particle size.
  • Validation: The process of establishing documented evidence that a system or process meets its intended requirements.
  • Criticality: The level of impact that the equipment has on product quality.

Roles

  • Validation Team: Responsible for executing the validation plan and documenting results.
  • Quality Assurance: Ensures compliance with regulatory requirements and approves validation documentation.
  • Production Personnel: Operate the equipment and provide feedback on performance during validation.

Lifecycle Procedure

  1. Conduct a User Requirement Specification (URS) review.
  2. Perform Design Qualification (DQ) to verify design meets requirements.
  3. Complete Installation Qualification (IQ) to ensure equipment is installed correctly.
  4. Execute Operational Qualification (OQ) to confirm equipment operates within specified limits.
  5. Finalize Performance Qualification (PQ) to demonstrate equipment produces acceptable results under normal operating conditions.

Good Documentation Practices (GDP) Controls

All validation documentation must be completed in accordance with Good Documentation Practices. This includes ensuring documents are signed, dated, and stored in a controlled manner.

Acceptance Criteria Governance

Acceptance criteria will be established based on the User Requirement Specification (URS) and must be met for each phase of validation (DQ, IQ, OQ, PQ).

Calibration and Preventive Maintenance (PM) Governance

The Jet Mill / Micronizer must undergo regular calibration and preventive maintenance as per the manufacturer’s recommendations and internal policies to ensure continued performance and compliance.

Change Control Triggers

Any changes to the equipment, processes, or materials that may impact the performance or validation status of the Jet Mill / Micronizer must initiate a change control process.

Revalidation Triggers and Periodic Review

Revalidation of the Jet Mill / Micronizer is required every 24 months or upon significant change to the equipment, process, or product.
