Objective
The purpose of this Operational Qualification (OQ) protocol is to verify that the Jet Mill / Micronizer operates within specified parameters to ensure product quality during the micronization process.
Scope
This protocol applies to the Jet Mill / Micronizer located in the Production/Milling area, and it covers the operational qualification of the equipment used for micronization of solid dosage forms.
Responsibilities
The validation team is responsible for executing this protocol, documenting results, and ensuring compliance with the acceptance criteria. The Quality Assurance team will review and approve the protocol.
Prerequisites
All personnel involved in the execution of this protocol must be trained on the operation of the Jet Mill / Micronizer and familiar with the relevant Standard Operating Procedures (SOPs).
Equipment Description
The Jet Mill / Micronizer is designed for the micronization of solid dosage forms using high-velocity air jets to achieve the desired particle size distribution (PSD). It operates under controlled air pressure and feed rate conditions.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| OQ-001 | Verify air pressure settings | Air pressure maintained within specified range | Calibration records |
| OQ-002 | Measure feed rate | Feed rate within defined limits | Feed rate logs |
| OQ-003 | Assess output PSD | Output PSD within acceptance criteria as per URS | PSD analysis report |
Detailed Test Cases
Test Case OQ-001: Air Pressure Verification
1. Set the Jet Mill / Micronizer to the specified air pressure setting.
2. Use calibrated pressure gauges to measure the air pressure.
3. Record the measurements and verify against the acceptance criteria.
Test Case OQ-002: Feed Rate Measurement
1. Set the feed mechanism to the specified rate.
2. Measure the actual feed rate using a calibrated scale.
3. Document the feed rate and compare with acceptance criteria.
Test Case OQ-003: Output PSD Assessment
1. Collect samples of the output material after micronization.
2. Analyze the particle size distribution using a laser diffraction method.
3. Compare results against the acceptance criteria defined in the URS.
Deviations
Any deviations from the protocol must be documented and justified. A deviation report must be generated for review and approval by Quality Assurance.
