Objective
The objective of this Installation Qualification (IQ) protocol is to verify that the Sigma Mixer/Kneader is installed according to the manufacturer’s specifications and is functioning as intended for high-viscosity mass mixing in solid dosage form production.
Scope
This protocol applies to the Sigma Mixer/Kneader located in the Production/Granulation area and is intended for use in the manufacturing of solid dosage forms.
Responsibilities
The Validation Team is responsible for executing this protocol, documenting results, and ensuring compliance with regulatory requirements.
Prerequisites
1. Installation of Sigma Mixer/Kneader completed.
2. All relevant personnel trained on equipment operation.
3. Availability of necessary tools and materials for testing.
Equipment Description
The Sigma Mixer/Kneader is designed for high-viscosity mass mixing, primarily used in the production of solid dosage forms. It features adjustable blade speed, temperature control, and vacuum capabilities to ensure effective mixing.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| IQ-001-A | Verify blade speed settings. | Blade speed must be within specified range as per URS. | Calibration report, logbook entries. |
| IQ-001-B | Check temperature control functionality. | Temperature must remain stable within specified limits. | Temperature logs. |
| IQ-001-C | Assess vacuum functionality. | Vacuum levels must meet operational specifications. | Vacuum gauge readings. |
Detailed Test Cases
Test Case 1: Blade Speed Verification
Procedure: Set the blade speed to the specified RPM and record the actual speed.
Acceptance Criteria: Blade speed must be within the range specified in the URS.
Evidence: Calibration report showing blade speed measurement.
Test Case 2: Temperature Control Assessment
Procedure: Set the mixer to the desired temperature and monitor for stability over 30 minutes.
Acceptance Criteria: Temperature must remain stable within ±2°C of the set point.
Evidence: Temperature log showing readings over the test period.
Test Case 3: Vacuum Functionality Check
Procedure: Engage the vacuum system and record the vacuum level.
Acceptance Criteria: Vacuum must reach and maintain the specified level as per operational requirements.
Evidence: Vacuum gauge readings documented during the test.
Deviations
Any deviations from the acceptance criteria must be documented and investigated. Appropriate corrective actions should be taken, and re-testing should be performed as necessary.
Approvals
This protocol must be approved by the Validation Manager and the Production Manager before execution.
