Operational Qualification Protocol for Continuous Blender
Meta Description: This document outlines the Operational Qualification protocol for the Continuous Blender used in Solid Dosage Form production, ensuring compliance with regulatory standards.
Tags: Equipment Validation, OQ, Continuous Blender, Solid Dosage Form, Production, Quality Control
Objective
The objective of this Operational Qualification (OQ) protocol is to verify that the Continuous Blender operates within specified limits and meets the requirements as defined in the User Requirements Specification (URS) Annex 11.
Scope
This protocol applies to the Continuous Blender used in the production of Solid Dosage Forms (OSD) within the Production/Continuous Manufacturing area. It encompasses all critical parameters affecting the quality of the product.
Responsibilities
- Validation Team: Responsible for executing the OQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the protocol and results.
- Production Team: Responsible for providing access to the equipment and relevant operational data.
Prerequisites
- Completion of Installation Qualification (IQ) for Continuous Blender.
- Availability of all necessary equipment and materials for testing.
- Training of personnel on the operation of the Continuous Blender.
Equipment Description
The Continuous Blender is designed for continuous blending of solid dosage forms, ensuring uniformity and consistency in the final product. The critical parameters include feed rates, RTD, RPM, and PLC controls, which directly impact the blending process.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| OQ-CB-01 | Verify feed rates at specified settings. | Feed rates within ±5% of target values. | Calibration logs, measurement records. |
| OQ-CB-02 | Check RTD readings during operation. | RTD readings within specified limits. | RTD calibration certificates, operation logs. |
| OQ-CB-03 | Test RPM settings against PLC controls. | RPM within ±3% of setpoint. | PLC logs, RPM measurement records. |
Detailed Test Cases
Test Case OQ-CB-01
Objective: To verify that the feed rates are within acceptable limits.
Procedure: Set the feed rate to the specified target and measure the actual feed rate using calibrated scales.
Acceptance Criteria: Feed rates must be within ±5% of target values.
Expected Outcome: Documented feed rates must meet acceptance criteria.
Test Case OQ-CB-02
Objective: To check RTD readings during blending operations.
Procedure: Monitor RTD readings throughout the blending process and compare with specified limits.
Acceptance Criteria: RTD readings must remain within defined limits.
Expected Outcome: RTD logs must confirm compliance.
Test Case OQ-CB-03
Objective: To validate RPM settings against PLC controls.
Procedure: Adjust RPM settings via PLC and measure actual RPM using a tachometer.
Acceptance Criteria: RPM must be within ±3% of the setpoint.
Expected Outcome: RPM measurements must align with PLC settings.
Deviations
Any deviations from the acceptance criteria must be documented and assessed for impact on product quality. Corrective actions should be identified and implemented as necessary.
