Design Qualification Protocol for Cartoning Machine in Solid Dosage Form Validation

Objective: To establish a Design Qualification (DQ) protocol for the validation of the Cartoning Machine used in the packaging of solid dosage forms, ensuring compliance with regulatory requirements and operational specifications.

Scope: This protocol applies to the Cartoning Machine utilized in the Packaging/Secondary area for carton packing of solid dosage forms, with a direct impact on product quality and compliance.

Responsibilities:

  • Validation Team: Responsible for executing the DQ protocol and documenting results.
  • Quality Assurance: Ensures compliance with regulatory standards and reviews the protocol.
  • Engineering: Provides technical support for the Cartoning Machine.

Prerequisites:

  • Completion of User Requirement Specification (URS) for the Cartoning Machine.
  • Installation Qualification (IQ) of the Cartoning Machine.
  • Availability of relevant documentation (manuals, specifications, etc.).

Equipment Description: The Cartoning Machine is designed for the efficient packing of solid dosage forms into cartons, with features including leaflet detection, reject logic, and audit trail capabilities. It operates within the Packaging/Secondary area and is classified as a major criticality equipment impacting product quality directly.

Test ID Procedure Acceptance Criteria Evidence
DQ-001 Verify leaflet detection functionality. Leaflet detection must operate with 98% accuracy. Test report and system logs.
DQ-002 Evaluate reject logic performance. Reject logic must activate within 2 seconds of detection. Performance test results.
DQ-003 Audit trail verification. Audit trail must capture all operational events. Audit trail report.

Detailed Test Cases:

  • Test Case DQ-001: Conduct a test to verify the accuracy of leaflet detection by running 100 cartons through the machine and recording detection accuracy.
  • Test Case DQ-002: Measure the time taken for the reject logic to activate after a faulty leaflet is detected by simulating faulty leaflet insertion.
  • Test Case DQ-003: Review the audit trail logs for completeness and accuracy after a full operational cycle of the machine.

Deviations: Any deviations from the acceptance criteria will be documented and addressed according to the deviation management procedure.
