Installation Qualification Protocol for Depyrogenation Tunnel in Ophthalmics Production

Objective: To establish and document the installation qualification of the Depyrogenation Tunnel used for sterilizing and depyrogenating glass bottles for sterile ophthalmic products.

Scope: This protocol applies to the Installation Qualification of the Depyrogenation Tunnel in the Production area for the manufacturing of sterile eye drops and eye ointments.

Responsibilities:

  • Validation Team: Oversee and execute the IQ protocol.
  • Quality Assurance: Review and approve the protocol and results.
  • Production Team: Provide operational support during testing.

Prerequisites:

  • Completion of equipment installation as per manufacturer specifications.
  • Training of personnel on equipment operation.
  • Availability of necessary documentation, including URS, Annex 15, and Annex 11.

Equipment Description:

The Depyrogenation Tunnel is designed to sterilize and depyrogenate glass bottles using a dry heat process. Key critical parameters include temperature profile, belt speed, dwell time, and endotoxin reduction.

Test ID Procedure Acceptance Criteria Evidence
IQ-01 Verify installation against manufacturer’s specifications. All components installed as per specifications. Installation checklist signed by validation team.
IQ-02 Check calibration of temperature sensors. Calibration within specified limits. Calibration certificates.
IQ-03 Verify belt speed and dwell time settings. Belt speed and dwell time meet defined parameters. Operational logs.
IQ-04 Conduct endotoxin reduction test. Endotoxin levels reduced to acceptable limits. Test results report.

Detailed Test Cases:

  • Test Case IQ-01: Inspect the installation of the Depyrogenation Tunnel components, ensuring all parts match the manufacturer’s installation guide.
  • Test Case IQ-02: Perform calibration checks on temperature sensors using calibrated equipment. Document results.
  • Test Case IQ-03: Measure belt speed and dwell time using a stopwatch and confirm settings against operational parameters.
  • Test Case IQ-04: Use validated methods to test endotoxin levels before and after the depyrogenation process.

Deviations:

Any deviations from the acceptance criteria must be documented and investigated. Corrective actions should be implemented and re-testing performed as necessary.
