Performance Qualification Protocol for Inline Emulsification System

Objective: To validate the performance of the Inline Emulsification System used for forming emulsion droplets in the production of microspheres and microcapsules.

Scope: This protocol applies to the Emulsification System utilized in the R&D/Production area for the formulation of microspheres. It encompasses all critical parameters affecting product quality.

Responsibilities:

  • Validation Team: Prepare and execute the PQ protocol.
  • Quality Assurance: Review and approve the protocol and results.
  • Production Team: Provide necessary data and support during testing.

Prerequisites:

  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
  • Availability of all necessary equipment and materials.
  • Training of personnel on equipment operation and data recording.

Equipment Description:
The Inline Emulsification System is designed to produce uniform emulsion droplets for microspheres. It operates under controlled shear rates, flow temperatures, and pressures to ensure product consistency and quality.

Test ID Procedure Acceptance Criteria Evidence
PQ-001 Measure shear rate during operation. Shear rate within specified range as per URS Annex11. Shear rate log records.
PQ-002 Monitor flow temperature throughout the process. Flow temperature maintained within specified limits. Temperature records.
PQ-003 Record pressure during emulsification. Pressure readings within acceptable range. Pressure log records.

Detailed Test Cases:

  1. Test Case ID: PQ-001
    • Procedure: Record shear rate during emulsification for three consecutive runs.
    • Expected Outcome: Shear rate remains consistent across all runs.
  2. Test Case ID: PQ-002
    • Procedure: Monitor and log flow temperature at 5-minute intervals during operation.
    • Expected Outcome: Temperature remains within specified limits at all times.
  3. Test Case ID: PQ-003
    • Procedure: Record pressure at the start, middle, and end of the emulsification process.
    • Expected Outcome: Pressure readings are consistent and fall within the acceptable range.
