Installation Qualification Protocol for Liposome Extruder

This document outlines the Installation Qualification (IQ) protocol for the Extrusion System utilized in the production of Liposomes and Lipid Nanoparticles (LNP).

Objective: To verify that the Extrusion System is installed correctly and operates according to specified requirements.

Scope: This protocol applies to the Extrusion System located in the R&D/Production area, ensuring compliance with URS Annex11.

Responsibilities:

  • Validation Team: Conduct IQ testing and documentation.
  • Quality Assurance: Review and approve the IQ protocol.
  • Maintenance Team: Ensure equipment is installed and maintained properly.

Prerequisites:

  • Completion of equipment installation.
  • Availability of all necessary documentation (URS, equipment manuals, etc.).
  • Training of personnel on equipment operation and validation procedures.

Equipment Description:

The Extrusion System (Liposome Extruder) is designed for size control of liposomes and lipid nanoparticles via membrane extrusion. The system is critical for ensuring product quality and consistency.

Test ID Procedure Acceptance Criteria Evidence
IQ-01 Verify installation against equipment specifications. All components are installed as per specifications. Installation checklist signed off.
IQ-02 Check calibration of pressure gauge. Pressure gauge within ±5% of standard. Calibration certificate.
IQ-03 Verify membrane pore size. Pore size matches specifications. Membrane specifications document.
IQ-04 Record operational temperature and flow rates. Temperature and flow rates within specified limits. Operational logs.

Detailed Test Cases:

  • Test Case IQ-01: Inspect all installation components for conformity to specifications.
  • Test Case IQ-02: Conduct calibration checks on the pressure gauge and document results.
  • Test Case IQ-03: Measure membrane pore size using appropriate analytical techniques.
  • Test Case IQ-04: Monitor and record temperature and flow rates during initial operation.

Deviations:

Any deviations from acceptance criteria must be documented and assessed. A corrective action plan will be developed and implemented as necessary.
