Operational Qualification Protocol for Fluid Bed Dryer in Polymeric Nanoparticle Processing
Objective: The objective of this Operational Qualification (OQ) is to validate the performance of the Fluid Bed Dryer used for the drying and granulation of polymeric nanoparticles, ensuring compliance with defined acceptance criteria.
Scope: This protocol applies to the Fluid Bed Dryer utilized in the R&D and Production areas for the processing of polymeric nanoparticles such as PLGA.
Responsibilities:
- Validation Team: Responsible for executing the OQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the OQ protocol and results.
- Equipment Operator: Responsible for operating the Fluid Bed Dryer as per the defined procedures.
Prerequisites:
- Completion of Installation Qualification (IQ) for the Fluid Bed Dryer.
- Training of personnel on the operation of the Fluid Bed Dryer.
- Availability of necessary materials for testing.
Equipment Description: The Fluid Bed Dryer is designed for the drying and granulation of particles, utilizing controlled airflow and temperature to achieve desired moisture content and particle characteristics. It is critical for the processing of polymeric nanoparticles.
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| OQ-001-A | Verify Airflow Temperature | Airflow temperature within specified range (e.g., 40-60°C) | Temperature log records |
| OQ-001-B | Verify Product Temperature | Product temperature within specified range (e.g., 30-50°C) | Product temperature log records |
| OQ-001-C | Record Keeping Verification | All records maintained accurately and retrievable | Audit of logbooks |
Detailed Test Cases:
Test Case OQ-001-A:
- Procedure: Set the Fluid Bed Dryer to the specified airflow temperature. Monitor and record the temperature for 30 minutes.
- Acceptance Criteria: The airflow temperature must remain within the defined range of 40-60°C.
- Evidence: Temperature log records must be printed and signed by the operator.
Test Case OQ-001-B:
- Procedure: Load the product into the Fluid Bed Dryer and monitor the product temperature for 30 minutes.
- Acceptance Criteria: The product temperature must remain within the defined range of 30-50°C.
- Evidence: Product temperature log records must be printed and signed by the operator.
Test Case OQ-001-C:
- Procedure: Review the logbooks and electronic records for completeness and accuracy.
- Acceptance Criteria: All records must be maintained accurately and be retrievable.
- Evidence: Audit report of logbooks and electronic records.
Deviations:
Any deviations from the outlined procedures or acceptance criteria must be documented in a deviation report and reviewed by the Quality Assurance team.
Approvals:
All results must be reviewed and approved by the Validation Team and Quality Assurance prior to finalization of the OQ protocol.
