Objective

The purpose of this Design Qualification (DQ) Protocol is to ensure that the High Shear Mixer / Homogenizer is properly designed, installed, and operates in accordance with the specified requirements for the production of transdermal patches.

Scope

This protocol applies to the High Shear Mixer / Homogenizer utilized in the production area for the manufacturing of transdermal patches, focusing on the uniform distribution of the drug within the adhesive matrix.

Responsibilities

  • Validation Team: Responsible for the execution and documentation of the DQ protocol.
  • Quality Assurance: Responsible for reviewing and approving the DQ protocol and results.
  • Production Team: Responsible for operating the equipment as per the validated procedures.

Prerequisites

  • Completion of User Requirement Specifications (URS) as per Annex 11.
  • Installation Qualification (IQ) completed and documented.
  • Personnel trained on equipment operation and safety protocols.

Equipment Description

The High Shear Mixer / Homogenizer is designed to ensure uniform drug distribution within the adhesive matrix of transdermal patches. Key specifications include adjustable speed settings, temperature control, and viscosity measurement capabilities. The equipment is equipped with PLC logging for data integrity.

Test Plan

Test ID Procedure Acceptance Criteria Evidence
TDS-DQ-001 Verify speed settings during operation. Speed must be within specified range as per URS. PLC log report.
TDS-DQ-002 Measure shear temperature during operation. Temperature must remain within defined limits. Temperature log report.
TDS-DQ-003 Check viscosity of the adhesive matrix. Viscosity must be within specified range. Viscosity measurement report.

Detailed Test Cases

Test Case: Speed Verification

Procedure:

  1. Set the mixer to the specified speed.
  2. Record the speed using the PLC log.
  3. Compare the recorded speed with the acceptance criteria.

Expected Result: Speed recorded must match the specified range.

Test Case: Shear Temperature Measurement

Procedure:

  1. Run the mixer for a defined period.
  2. Monitor and record the shear temperature.
  3. Ensure the temperature is within the defined limits.

Expected Result: Temperature must remain within acceptance criteria.

Test Case: Viscosity Check

Procedure:

  1. After mixing, take a sample of the adhesive matrix.
  2. Measure the viscosity using a viscometer.
  3. Compare the measurement with the specified range.

Expected Result: Viscosity must be within the acceptance range.

Deviations

Any deviations from the acceptance criteria must be documented and investigated. A root cause analysis should be conducted, and corrective actions must be implemented to prevent recurrence.

Approvals

This Design Qualification Protocol must be reviewed and approved by the Quality Assurance department prior to execution.
