Objective

The objective of this Performance Qualification (PQ) protocol is to ensure that the High Shear Mixer / Homogenizer consistently produces transdermal patches with uniform drug distribution in the adhesive matrix, thereby meeting the defined acceptance criteria.

Scope

This protocol applies to the High Shear Mixer / Homogenizer used in the production area for the manufacture of transdermal patches (TDS). It covers critical parameters, acceptance criteria, and responsibilities involved in the qualification process.

Responsibilities

  • Validation Team: Responsible for the execution and documentation of the PQ protocol.
  • Production Team: Ensure equipment is maintained and operated according to the standard operating procedures.
  • Quality Assurance: Review and approve the protocol and results.

Prerequisites

  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
  • Availability of necessary materials and equipment.
  • Training of personnel on the use of the High Shear Mixer / Homogenizer.

Equipment Description

The High Shear Mixer / Homogenizer is designed to ensure uniform mixing of the adhesive matrix for transdermal patches. It operates under controlled speed, shear, temperature, and viscosity conditions and is equipped with PLC for data logging.

Test Plan

Test ID Procedure Acceptance Criteria Evidence
PQ-001 Measure speed during operation Speed within specified range PLC logs
PQ-002 Measure shear during operation Shear within specified range PLC logs
PQ-003 Measure temperature during operation Temperature within specified range PLC logs
PQ-004 Measure viscosity of the mixture Viscosity within specified range Viscosity test reports

Detailed Test Cases

Test Case 1: Speed Measurement

  • Procedure: Operate the mixer at defined speeds and record the actual speed using the PLC.
  • Acceptance Criteria: Speed must be within the specified range of [insert range].
  • Evidence: PLC logs will be reviewed to confirm speed compliance.

Test Case 2: Shear Measurement

  • Procedure: Monitor shear rates during operation.
  • Acceptance Criteria: Shear must be within the specified range of [insert range].
  • Evidence: PLC logs will be reviewed to confirm shear compliance.

Test Case 3: Temperature Measurement

  • Procedure: Monitor temperature during the mixing process.
  • Acceptance Criteria: Temperature must be within the specified range of [insert range].
  • Evidence: PLC logs will be reviewed to confirm temperature compliance.

Test Case 4: Viscosity Measurement

  • Procedure: Measure viscosity of the final mixture using a viscometer.
  • Acceptance Criteria: Viscosity must be within the specified range of [insert range].
  • Evidence: Viscosity test reports will be reviewed.

Deviations

Any deviations from the defined acceptance criteria must be documented and assessed for impact on product quality. A deviation report must be generated and reviewed by the Quality Assurance team.
