Objective
The objective of this Installation Qualification (IQ) protocol is to verify that the Lipid Melting / Lipid Preparation Vessel is installed correctly and operates according to the specified requirements for the preparation of lipid phase.
Scope
This protocol applies to the Lipid Melting / Lipid Preparation Vessel (Jacketed SS/Glass) used in the R&D and Production areas for the preparation of lipid phase in the formulation of liposomes and lipid nanoparticles (LNP).
Responsibilities
- Validation Team: Responsible for executing the IQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the IQ documentation.
- Maintenance Team: Responsible for ensuring the equipment is maintained and calibrated prior to IQ execution.
Prerequisites
- Installation of Lipid Melting / Lipid Preparation Vessel completed.
- All relevant documentation (URS, specifications, etc.) available for review.
- Calibration of the equipment completed and documented.
Equipment Description
The Lipid Melting / Lipid Preparation Vessel is a jacketed stainless steel/glass vessel designed for the preparation of lipid phase. It features precise temperature control, mixing capabilities, and is equipped with data logging for critical parameters.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| IQ-01 | Verify temperature control functionality. | Temperature within ±1°C of setpoint. | Calibration certificate and temperature logs. |
| IQ-02 | Verify mixing speed accuracy. | Mixing speed within ±10% of setpoint. | Calibration certificate and mixing speed logs. |
| IQ-03 | Verify hold time capability. | Hold time accurately maintained as per specifications. | Hold time logs and temperature records. |
Detailed Test Cases
Test Case: IQ-01 – Temperature Control Functionality
Procedure: Set the temperature to the specified setpoint and monitor for stability.
Acceptance Criteria: Temperature must remain within ±1°C of setpoint for a minimum of 30 minutes.
Evidence: Document temperature readings and stability logs.
Test Case: IQ-02 – Mixing Speed Accuracy
Procedure: Set the mixing speed and measure actual speed using calibrated equipment.
Acceptance Criteria: Mixing speed must be within ±10% of the setpoint.
Evidence: Documented mixing speed measurements.
Test Case: IQ-03 – Hold Time Capability
Procedure: Set the vessel to the required temperature and monitor for the specified hold time.
Acceptance Criteria: Hold time must be accurately maintained.
Evidence: Documented hold time records and temperature logs.
Deviations
Any deviations from the protocol must be documented and assessed for impact on validation. A deviation report must be generated and approved by Quality Assurance.
