Operational Qualification Protocol for Lyophilizer Vacuum System

Objective: To validate the Lyophilizer Vacuum System to ensure it maintains the required vacuum conditions for the production of sterile powders and lyophilized products.

Scope: This protocol applies to the Lyophilizer Vacuum System located in the production area, directly impacting product quality and compliance with regulatory standards.

Responsibilities:

  • Validation Team: Execute the OQ protocol and document results.
  • Quality Assurance: Review and approve the protocol and results.
  • Production Personnel: Ensure equipment is operational prior to testing.

Prerequisites:

  • Installation Qualification (IQ) must be completed and approved.
  • All necessary training for personnel involved must be completed.

Equipment Description:

The Lyophilizer Vacuum System is designed to maintain the required vacuum conditions essential for the lyophilization process of sterile powders. It is equipped with vacuum pressure integrity cycle logs to monitor performance.

Test ID Procedure Acceptance Criteria Evidence
OQ-01 Verify vacuum pressure integrity during a complete cycle. Vacuum pressure must remain within specified limits as per URS Annex11. Cycle log report.
OQ-02 Check for alarms and alerts during operation. No critical alerts should occur during the test. Alarm log report.
OQ-03 Perform data integrity checks on cycle logs. Data must be complete and without discrepancies. Data integrity report.

Detailed Test Cases:

  • Test Case OQ-01: Document the vacuum pressure readings at the start, during, and at the end of a complete cycle. Compare results against URS Annex11 specifications.
  • Test Case OQ-02: Monitor the system for any alarms or alerts during operation. Record the occurrence and response to any alerts.
  • Test Case OQ-03: Review cycle logs for completeness and accuracy. Verify that all data entries are timestamped and correspond with the operational timeline.

Deviations: Any deviations from the acceptance criteria must be documented and investigated. A corrective action plan should be developed and implemented.
