Operational Qualification Protocol for Nano Tracking Analysis System

Objective: The purpose of this Operational Qualification (OQ) protocol is to verify that the Nano Tracking Analysis (NTA) System operates according to the specified requirements and produces accurate and reliable data for particle concentration and size analysis.

Scope: This protocol applies to the Nano Tracking Analysis System used in the Quality Control (QC) and Research & Development (R&D) areas for critical NDDS analytics.

Responsibilities:

  • Validation Team: Responsible for executing the OQ protocol and documenting results.
  • Quality Assurance: Responsible for reviewing and approving the OQ protocol and results.
  • Equipment Owner: Responsible for ensuring the equipment is maintained and calibrated.

Prerequisites:

  • Installation Qualification (IQ) must be completed and approved.
  • All necessary training for personnel on the NTA System must be completed.
  • Documentation of maintenance and calibration must be available.

Equipment Description:

The Nano Tracking Analysis (NTA) System is an advanced instrument used for measuring particle concentration and size in nanoparticle formulations. It provides critical data for the development and quality control of drug delivery systems.

Test ID Procedure Acceptance Criteria Evidence
OQ-001 Verify system calibration using standard reference particles. Calibration must be within ±5% of the nominal value. Calibration report.
OQ-002 Conduct accuracy checks using known particle sizes. Measured sizes must match known sizes within ±10%. Accuracy check report.
OQ-003 Perform data integrity audit trail check. Audit trail must show complete data recording with no gaps. Audit trail report.

Detailed Test Cases

Test Case OQ-001

Objective: To verify the calibration of the NTA system.

Procedure: Use standard reference particles to calibrate the system and record the results.

Expected Outcome: Calibration values should be within ±5% of the nominal value.

Test Case OQ-002

Objective: To check the accuracy of particle size measurements.

Procedure: Measure known particle sizes and compare with expected values.

Expected Outcome: Measured sizes must match known sizes within ±10%.

Test Case OQ-003

Objective: To ensure data integrity through audit trail verification.

Procedure: Review the audit trail for completeness and accuracy.

Expected Outcome: The audit trail must show complete data recording with no gaps.

Deviations

Any deviations from the established acceptance criteria must be documented and investigated. A root cause analysis should be performed, and corrective actions should be implemented as necessary.

Approvals

All test results must be reviewed and approved by the Quality Assurance team before the NTA system is deemed operationally qualified.
