Performance Qualification Protocol for Analytical Weighing Balance in API Production
Meta Description: This document outlines the Performance Qualification Protocol for an Analytical Weighing Balance used in API manufacturing, focusing on critical parameters and acceptance criteria.
Tags: Performance Qualification, Weighing Balance, API Manufacturing, Equipment Validation
Objective
The objective of this protocol is to establish the performance qualification requirements for the Analytical Weighing Balance used in the production and dispensing of Active Pharmaceutical Ingredients (APIs).
Scope
This protocol applies to the Analytical Weighing Balance utilized in the API manufacturing area, focusing on its capability to accurately weigh API materials critical to product quality.
Responsibilities
- Validation Team: Responsible for executing the qualification protocol and documenting results.
- Quality Assurance (QA): Responsible for reviewing and approving the protocol and results.
- Production Personnel: Responsible for operating the weighing balance and ensuring compliance with the protocol during use.
Prerequisites
- Completion of Installation Qualification (IQ) for the weighing balance.
- Calibration of the weighing balance per URS SOP Calibration.
- Training of personnel on the operation of the weighing balance.
Equipment Description
The Analytical Weighing Balance is designed for precise measurement of small quantities of materials, crucial for the accurate formulation of APIs. It is equipped with features to ensure high accuracy and repeatability.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| PQ-01 | Accuracy Test | Within ±0.1% of standard weight | Calibration report |
| PQ-02 | Repeatability Test | Standard deviation < 0.05g over 10 measurements | Measurement log |
| PQ-03 | Linearity Test | Deviation < 0.1% across range | Test report |
Detailed Test Cases
Test Case 1: Accuracy Test
Procedure: Weigh a known standard weight (e.g., 100g) and record the measurement. Repeat for three trials.
Acceptance Criteria: The measured weight must be within ±0.1% of the standard weight.
Evidence: Documented results in the calibration report.
Test Case 2: Repeatability Test
Procedure: Weigh the same sample ten times and record the results.
Acceptance Criteria: The standard deviation of the ten measurements must be less than 0.05g.
Evidence: Measurement log documenting all ten results.
Test Case 3: Linearity Test
Procedure: Weigh standard weights across the balance’s range (e.g., 10g, 50g, 100g) and record the measurements.
Acceptance Criteria: Deviation from the expected value must be less than 0.1% across the range.
Evidence: Test report summarizing the results and deviations.
Deviations
Any deviations from the acceptance criteria must be documented, investigated, and resolved before the qualification can be considered complete. A corrective action plan should be developed if necessary.
