Objective

The objective of this Performance Qualification (PQ) protocol is to validate the Zeta Potential Analyzer for its intended use in measuring surface charge as part of the Critical Quality Attributes (CQA) and Critical Process Parameters (CPP) in the development of Novel Drug Delivery Systems (NDDS).

Scope

This protocol applies to the Zeta Potential Analyzer used in the Quality Control (QC) and Research & Development (R&D) departments for critical NDDS analytics.

Responsibilities

The following personnel are responsible for executing this protocol:

  • Validation Team: Oversee the execution of the PQ protocol.
  • Quality Control Personnel: Conduct the tests as per the protocol.
  • Quality Assurance: Review and approve the PQ protocol and results.

Prerequisites

  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
  • Training of personnel on the use of the Zeta Potential Analyzer.
  • Availability of necessary reagents and materials for testing.

Equipment Description

The Zeta Potential Analyzer is an instrument used to measure the surface charge of particles in suspension, providing critical data for the formulation and stability of NDDS. It is equipped with precision sensors to ensure accurate readings.

Test Plan

Test ID Procedure Acceptance Evidence
TP-01 Verify accuracy of surface charge measurements. Measurements within ±5% of known standards. Calibration report and measurement data.
TP-02 Assess repeatability of measurements. Standard deviation < 2% across replicate measurements. Statistical analysis report.
TP-03 Verify audit trail functionality. Complete audit trail for all measurements. Audit trail report.

Detailed Test Cases

Test Case 1: Accuracy of Surface Charge Measurements

Procedure: Measure the zeta potential of a standard solution. Compare results to known values.

Acceptance Criteria: Measurements must be within ±5% of known standards.

Data Integrity Checks: Ensure all measurements are logged in the system with timestamps and user IDs.

Test Case 2: Repeatability of Measurements

Procedure: Conduct multiple measurements of the same sample under identical conditions.

Acceptance Criteria: Standard deviation must be < 2% across replicate measurements.

Data Integrity Checks: Verify that all replicate measurements are recorded and linked to the same sample ID.

Test Case 3: Audit Trail Verification

Procedure: Review the audit trail for the Zeta Potential Analyzer.

Acceptance Criteria: A complete audit trail must be available for all measurements conducted.

Data Integrity Checks: Confirm that all entries in the audit trail are tamper-proof and include user information.

Deviations

Any deviations from this protocol must be documented in a deviation report and approved by Quality Assurance. Corrective actions must be taken as necessary.
