Objective
The objective of this Installation Qualification (IQ) protocol is to verify that the Ultrafiltration/Diafiltration (UF/DF) skid is installed according to the manufacturer’s specifications and operational requirements, ensuring its readiness for use in the purification and solvent exchange of NDDS products such as Liposomes and Lipid Nanoparticles.
Scope
This protocol applies to the installation of the Ultrafiltration/Diafiltration skid in the R&D/Production area, focusing on its use for direct product impact with criticality classified as critical.
Responsibilities
- Validation Team: Responsible for executing the IQ protocol and documenting results.
- Quality Assurance: Responsible for reviewing and approving the IQ protocol and results.
- Engineering: Responsible for providing technical support during installation and testing.
Prerequisites
- Completion of equipment installation by qualified personnel.
- Availability of installation manuals and manufacturer’s specifications.
- Training of personnel on equipment operation and safety procedures.
Equipment Description
The Ultrafiltration/Diafiltration (UF/DF) skid is designed for the purification and solvent exchange of NDDS products. It utilizes membrane technology to separate particles based on size, ensuring high efficiency and product integrity. The skid includes pressure flow monitoring and membrane integrity testing capabilities.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| IQ-1 | Verify installation against manufacturer’s specifications. | All components installed as per specifications. | Installation checklist signed by technician. |
| IQ-2 | Check pressure flow membrane integrity. | Pressure flow readings within acceptable limits. | Pressure flow test records. |
| IQ-3 | Review calibration of sensors and gauges. | All sensors calibrated and within tolerance. | Calibration certificates. |
Detailed Test Cases
Test Case: IQ-1
Objective: To ensure all components are installed as specified.
Procedure: Conduct a walkthrough of the installation with reference to the manufacturer’s installation manual.
Acceptance Criteria: All components should match the installation manual.
Evidence: Documented installation checklist signed off by the installation technician.
Test Case: IQ-2
Objective: To verify membrane integrity through pressure flow testing.
Procedure: Perform a pressure flow test and record the data.
Acceptance Criteria: Pressure readings must be within the specified range.
Evidence: Pressure flow test records maintained in the validation file.
Test Case: IQ-3
Objective: To confirm that all sensors and gauges are calibrated.
Procedure: Review calibration certificates for all sensors and gauges used in the skid.
Acceptance Criteria: Calibration certificates must be current and within specified tolerances.
Evidence: Copies of calibration certificates filed with the validation documents.
Deviations
Any deviations observed during the IQ process must be documented, including the nature of the deviation, the impact on equipment performance, and the corrective actions taken. All deviations require approval from the Quality Assurance department.
