Operational Qualification Protocol for Ultracentrifuge in NDDS Applications

Objective: To validate the operational performance of the Ultracentrifuge for particle separation and purification in the development of polymeric nanoparticles.

Scope: This protocol applies to the Ultracentrifuge used in the R&D/Production area for the direct impact on product quality in NDDS applications.

Responsibilities:

  • Validation Team: Overall execution and documentation of the OQ protocol.
  • Quality Assurance: Review and approval of the protocol and results.
  • Equipment Operator: Conducting the tests as per the protocol.

Prerequisites:

  • Training completed for equipment operation.
  • Equipment calibrated and maintained as per SOP.
  • All necessary materials and reagents available.

Equipment Description:

The Ultracentrifuge is designed for high-speed separation of particles based on density. It operates under controlled RPM, temperature, and time parameters, ensuring effective purification of polymeric nanoparticles.

Test ID Procedure Acceptance Criteria Evidence
OQ-001 Verify RPM settings at maximum capacity. RPM within ±10% of set point. Calibration log, RPM measurement data.
OQ-002 Check time accuracy during operation. Time deviation ≤ 5 seconds. Time tracking log.
OQ-003 Test temperature stability during operation. Temperature maintained within ±2°C of set point. Temperature log, equipment monitoring data.
OQ-004 Evaluate imbalance controls. No imbalance alarm triggered during operation. Imbalance test results.

Detailed Test Cases:

Test Case OQ-001: Run the Ultracentrifuge at maximum RPM for 10 minutes. Measure RPM using calibrated tachometer. Document RPM values and compare with set point.

Test Case OQ-002: Operate the Ultracentrifuge for a complete cycle and record time. Verify against the timer on the equipment.

Test Case OQ-003: Monitor temperature during a full operational cycle. Use calibrated thermometer to record temperature at intervals.

Test Case OQ-004: Conduct imbalance test by running the centrifuge with unevenly distributed samples. Check for alarms or errors during operation.

Deviations:

Any deviations from the acceptance criteria should be documented and investigated. A root cause analysis should be performed, and corrective actions documented.
