Objective

The objective of this Performance Qualification (PQ) protocol is to ensure that the Ultracentrifuge operates consistently and reliably under defined parameters for the separation and purification of Polymeric Nanoparticles.

Scope

This protocol applies to the Ultracentrifuge utilized in the R&D/Production area for the separation and purification of Polymeric Nanoparticles (e.g., PLGA).

Responsibilities

  • Validation Team: Responsible for executing the PQ protocol and documenting results.
  • Quality Assurance: Responsible for reviewing and approving the protocol and results.
  • Operators: Responsible for operating the equipment as per the validated procedures.

Prerequisites

  • Training on the operation of the Ultracentrifuge.
  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ).
  • Availability of necessary materials and resources for testing.

Equipment Description

The Ultracentrifuge is designed for high-speed particle separation and purification, capable of achieving various RPM settings, with precise control over time and temperature parameters. It is critical for the production of high-quality Polymeric Nanoparticles.

Detailed Test Cases

Test Case: PQ-UC-01

Objective: Verify RPM settings against the specified range.

Procedure: Set the Ultracentrifuge to the desired RPM and record the actual RPM displayed.

Acceptance Criteria: RPM must be within the specified range as per URS.

Evidence: Calibration certificate must be attached.

Test Case: PQ-UC-02

Objective: Measure operational time during particle separation.

Procedure: Start the Ultracentrifuge and record the time taken for separation.

Acceptance Criteria: Time must be within defined limits.

Evidence: Time log must be attached.

Objective: Monitor temperature during operation.

Procedure: Use a calibrated thermometer to measure temperature during operation.
