Design Qualification Protocol for Vacuum Emulsifier in Semi-Solid Production

Objective: To establish and document the Design Qualification (DQ) for the Vacuum Emulsifier utilized in the emulsification and mixing of creams, ointments, gels, and lotions.

Scope: This protocol applies to the Vacuum Emulsifier used in the Production/Manufacturing area for the direct impact on product quality in the production of semi-solids.

Responsibilities:

  • Validation Team: Responsible for executing the DQ protocol and documenting results.
  • Quality Assurance: Responsible for reviewing and approving the DQ protocol and results.
  • Production Team: Responsible for providing necessary operational insights and support during the DQ process.

Prerequisites:

  • Completion of User Requirements Specification (URS) for the Vacuum Emulsifier.
  • Installation Qualification (IQ) must be completed prior to DQ execution.

Equipment Description:

The Vacuum Emulsifier is a main manufacturing vessel designed for the emulsification and mixing of creams, ointments, gels, and lotions. It operates under controlled conditions of agitation speed, temperature, vacuum, and homogenization.

Test ID Procedure Acceptance Criteria Evidence
DQ-VE-001 Verify agitation speed settings Agitation speed must be within specified limits PLC logs
DQ-VE-002 Measure temperature during operation Temperature must be within specified limits Temperature logs
DQ-VE-003 Check vacuum levels Vacuum must meet operational standards Vacuum gauge readings
DQ-VE-004 Homogenization effectiveness test Homogenization must meet product specifications Sample analysis results

Detailed Test Cases:

  • Test Case 1: Verify that the agitation speed can be set to the required RPM and is maintained throughout the mixing process.
  • Test Case 2: Ensure that the temperature control system can accurately maintain the desired temperature range throughout the emulsification process.
  • Test Case 3: Confirm that the vacuum system is capable of achieving the necessary vacuum levels for effective emulsification.
  • Test Case 4: Assess the performance of the homogenization process through sample analysis to ensure product specifications are met.

Data Integrity Checks:

  • Regular audits of PLC logs to ensure all critical parameters are recorded accurately.
  • Cross-verification of temperature and vacuum readings with calibrated instruments.
  • Documentation of all deviations and corrective actions taken.

Deviations: Any deviations from the acceptance criteria must be documented, investigated, and resolved prior to final approval of the DQ protocol.
