Performance Qualification Protocol for CO2 Incubator in Upstream Processing

Meta Description: This document outlines the Performance Qualification protocol for the CO2 Incubator used in upstream processing for biologics, focusing on critical parameters and acceptance criteria.

Objective

To establish a Performance Qualification (PQ) protocol for the CO2 Incubator used in the growth of cell cultures, ensuring compliance with regulatory standards and product impact assessments.

Scope

This protocol applies to the CO2 Incubator utilized in the Production/Cell Culture area for biologics upstream processing. It encompasses all aspects of validation and performance testing.

Responsibilities

The Validation Team is responsible for executing the PQ protocol, documenting results, and ensuring compliance with the acceptance criteria. The Quality Assurance (QA) team will review and approve the protocol and results.

Prerequisites

All personnel involved must be trained in Good Manufacturing Practices (GMP) and have received specific training on the CO2 Incubator operation and validation protocols.

Equipment Description

The CO2 Incubator is designed for the growth of cell cultures, providing controlled temperature, humidity, and CO2 levels to optimize cell growth conditions. It is critical for biologics production as it directly impacts product quality.

Detailed Test Cases

Test Case TP-001: Verify Temperature Control

Procedure: Set the incubator to the desired temperature. Monitor the temperature for a period of 24 hours and record the PLC logs.

Acceptance Criteria: Temperature must remain within ± 1°C of the setpoint.

Evidence: PLC logs will be reviewed and documented.

Test Case TP-002: Verify CO2 Concentration

Procedure: Set the CO2 level to the desired concentration. Monitor the CO2 levels for a period of 24 hours and record the PLC logs.

Acceptance Criteria: CO2 levels must remain within ± 0.5% of the setpoint.

Evidence: PLC logs will be reviewed and documented.

Test Case TP-003: Verify Humidity Control

Procedure: Set the humidity level to the desired percentage. Monitor the humidity for a period of 24 hours and record the PLC logs.

Acceptance Criteria: Humidity must remain within ± 5% of the setpoint.

Evidence: PLC logs will be reviewed and documented.

Deviations

Any deviations from the acceptance criteria must be documented with a detailed explanation and corrective actions taken. A deviation report must be submitted to QA for review.

Approvals

All results must be reviewed and approved by the Validation Team Lead and the Quality Assurance Manager.

Data Integrity Checks

1. Regular backup of PLC logs to secure storage.

2. Access controls to ensure only authorized personnel can modify settings.

3. Audit trails for all changes made to the incubator settings.
