Operational Qualification Protocol for CO2 Incubator in Biologics Upstream Processing
Document ID: OQ-CO2-001
Version: 1.0
Effective Date: 2023-10-01
Review Date: 2024-10-01
Objective
The objective of this Operational Qualification (OQ) protocol is to verify that the CO2 incubator operates within specified limits for critical parameters that impact the growth of cell cultures in the biologics production area.
Scope
This protocol applies to the CO2 incubator utilized in the Production/Cell Culture area for the growth of cell cultures, directly impacting product quality. The OQ will ensure that the equipment meets the requirements outlined in the User Requirement Specification (URS) Annex 11.
Responsibilities
The following roles are responsible for the execution and approval of this protocol:
- Validation Engineer: Responsible for protocol execution and documentation.
- Quality Assurance: Responsible for review and approval of the protocol.
- Production Manager: Ensures that the equipment is available for testing.
Prerequisites
- Installation Qualification (IQ) must be completed and approved.
- All personnel involved must be trained on the operation of the CO2 incubator.
- Necessary tools and materials for testing must be available.
Equipment Description
The CO2 incubator is designed to provide a controlled environment for the growth of cell cultures. It maintains specific temperature, CO2 concentration, and humidity levels to support optimal cell growth.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| OQ-01 | Verify temperature control | Temperature maintained at 37°C ± 0.5°C | PLC logs, temperature records |
| OQ-02 | Verify CO2 concentration | CO2 concentration maintained at 5% ± 0.2% | PLC logs, CO2 sensor readings |
| OQ-03 | Verify humidity control | Humidity maintained at 85% ± 5% | PLC logs, humidity records |
Detailed Test Cases
Test Case OQ-01: Verify Temperature Control
Procedure: Monitor the temperature of the CO2 incubator using calibrated thermometers and PLC logs for a minimum of 24 hours.
Acceptance Criteria: The temperature must remain stable at 37°C ± 0.5°C throughout the test period.
Evidence: Documented temperature logs and PLC data.
Test Case OQ-02: Verify CO2 Concentration
Procedure: Measure the CO2 concentration using calibrated gas analyzers and record data over a 24-hour period.
Acceptance Criteria: CO2 levels must remain within 5% ± 0.2% during the test period.
Evidence: Documented CO2 sensor readings and PLC data.
Test Case OQ-03: Verify Humidity Control
Procedure: Monitor humidity levels using calibrated hygrometers and PLC logs for a minimum of 24 hours.
Acceptance Criteria: Humidity must remain stable at 85% ± 5% throughout the test period.
Evidence: Documented humidity logs and PLC data.
Deviations
Any deviations from the acceptance criteria must be documented and investigated. A corrective action plan must be developed and approved by Quality Assurance.
