Document Number: PQ-FBC-001

Version: 1.0

Effective Date: [Insert Date]

Objective

The purpose of this Performance Qualification (PQ) protocol is to ensure that the Fluid Bed Coater (Wurster/Bottom Spray) operates within defined parameters to produce consistent and high-quality pellet coatings in compliance with regulatory requirements.

Scope

This protocol applies to the Fluid Bed Coater used in the production area for the coating of solid dosage forms (pellets) and addresses the critical parameters that impact product quality.

Responsibilities

  • Validation Team: Responsible for execution and documentation of the PQ protocol.
  • Quality Assurance: Responsible for review and approval of the protocol and results.
  • Production Personnel: Responsible for providing support during the validation process.

Prerequisites

  • Completion of Installation Qualification (IQ) and Operational Qualification (OQ) for the Fluid Bed Coater.
  • Availability of all necessary materials and equipment for testing.
  • Training of personnel on the use of the Fluid Bed Coater.

Equipment Description

The Fluid Bed Coater (Wurster/Bottom Spray) is designed for coating pellets with a uniform layer of coating material. The equipment operates by suspending the pellets in a fluidized state and spraying the coating solution onto them.

Test Plan

Test ID Procedure Acceptance Criteria Evidence
TP-01 Verify airflow temperature during operation. Within specified limits as per URS Annex11. Temperature log records.
TP-02 Measure spray rate during coating. Consistent spray rate as per URS Annex11. Spray rate measurement logs.
TP-03 Check filter differential pressure (DP). DP within acceptable range as per URS Annex11. Filter DP records.
TP-04 Validate PLC functionality. PLC operates without errors during the process. PLC operation logs.

Detailed Test Cases

Test Case 1: Airflow Temperature

Procedure: Monitor the airflow temperature during the coating process. Record the temperature at regular intervals.

Acceptance Criteria: Airflow temperature must remain within the specified range as defined in URS Annex11.

Evidence: Temperature logs must be maintained and reviewed.

Test Case 2: Spray Rate

Procedure: Measure the spray rate during the coating process using calibrated equipment.

Acceptance Criteria: The spray rate must be consistent and within the defined limits in URS Annex11.

Evidence: Spray rate measurement logs must be documented.

Test Case 3: Filter DP

Procedure: Check the differential pressure across the filter before, during, and after the process.

Acceptance Criteria: The DP must remain within the acceptable range as specified in URS Annex11.

Evidence: Filter DP records must be reviewed and maintained.

Test Case 4: PLC Functionality

Procedure: Validate the PLC by running a test cycle and observing for any errors or alarms.

Acceptance Criteria: The PLC must operate without any errors throughout the process.

Evidence: PLC operation logs must be collected and reviewed.

Deviations

Any deviations from the established acceptance criteria must be documented in a deviation report and reviewed by the Quality Assurance team for impact assessment.
