Standard Operating Procedure for Equipment Validation of Glass-Lined Reactors in API Manufacturing
Meta Description: This SOP outlines the equipment validation process for Glass-Lined Reactors (GLR) used in chemical synthesis, ensuring compliance with regulatory standards and product quality.
Tags: Equipment Validation, API Manufacturing, Chemical Synthesis, Glass-Lined Reactor, SOP
Purpose
The purpose of this Standard Operating Procedure (SOP) is to establish a framework for the validation of Glass-Lined Reactors (GLR) used in the manufacturing of Active Pharmaceutical Ingredients (APIs) through chemical synthesis. This ensures that the equipment consistently produces products of the required quality.
Scope
This SOP applies to all Glass-Lined Reactors utilized in the production area for chemical synthesis within the organization. It encompasses all phases of the equipment validation lifecycle including Design Qualification (DQ), Installation Qualification (IQ), Operational Qualification (OQ), and Performance Qualification (PQ).
Definitions
- Validation: The process of establishing documented evidence that a system or process meets its intended requirements.
- Glass-Lined Reactor (GLR): A type of reactor used in chemical synthesis that features a glass lining to prevent corrosion and contamination.
- Criticality: The importance of the equipment in ensuring product quality.
- URS: User Requirements Specification.
Roles
- Validation Team: Responsible for executing the validation process and ensuring compliance with regulatory requirements.
- Quality Assurance: Oversees the validation process and ensures adherence to SOPs and regulatory standards.
- Production Personnel: Operate the GLR and provide input on operational performance.
Lifecycle Procedure
- Design Qualification (DQ): Ensure that the design of the GLR meets user requirements.
- Installation Qualification (IQ): Verify that the GLR is installed according to manufacturer specifications.
- Operational Qualification (OQ): Confirm that the GLR operates within specified limits under normal operating conditions.
- Performance Qualification (PQ): Demonstrate that the GLR consistently performs as intended during actual manufacturing processes.
Good Documentation Practices (GDP) Controls
All documentation related to the validation of the GLR must adhere to Good Documentation Practices (GDP), ensuring clarity, accuracy, and traceability throughout the validation lifecycle.
Acceptance Criteria Governance
Acceptance criteria for validation activities shall be established based on User Requirements Specifications (URS) and in compliance with Annex 11 and Annex 15 of the relevant regulatory guidelines.
Calibration and Preventive Maintenance Governance
The GLR must undergo regular calibration and preventive maintenance as per the manufacturer’s recommendations and internal policies, ensuring optimal performance and compliance with regulatory standards.
Change Control Triggers
Any changes affecting the GLR’s design, installation, operation, or maintenance must initiate a change control process to assess potential impacts on validation status.
Revalidation Triggers and Periodic Review
Revalidation of the GLR is required every 24 months or whenever significant changes occur that may impact equipment performance or product quality. A periodic review of the validation status shall also be conducted to ensure ongoing compliance.
