Objective
The objective of this protocol is to establish the Operational Qualification (OQ) of the Glass-Lined Reactor (GLR) used for chemical synthesis in API manufacturing, ensuring that the equipment operates within specified parameters and meets acceptance criteria.
Scope
This protocol applies to the Glass-Lined Reactor located in the Production/Reaction area, utilized for chemical synthesis reactions that have a direct impact on product quality.
Responsibilities
The Validation Team is responsible for the execution and documentation of the OQ protocol. The Quality Assurance team will review and approve the protocol and results.
Prerequisites
All personnel involved in the execution of this protocol must be trained in validation practices and familiar with the operation of the Glass-Lined Reactor. All necessary equipment and materials must be in place prior to execution.
Equipment Description
The Glass-Lined Reactor (GLR) is a critical piece of equipment used for chemical synthesis reactions. It is designed to handle high temperatures and pressures while maintaining product integrity. Key features include temperature control, pressure monitoring, agitation capabilities, and PLC logging for data collection.
Test Plan
| Test ID | Procedure | Acceptance Criteria | Evidence |
|---|---|---|---|
| OQ-GLR-001 | Verify temperature control functionality | Temperature within specified range | Temperature logs |
| OQ-GLR-002 | Verify pressure monitoring | Pressure within specified range | Pressure logs |
| OQ-GLR-003 | Verify agitation RPM | Agitation RPM within specified range | Agitation logs |
| OQ-GLR-004 | Review PLC logs for accuracy | Logs accurately reflect operations | PLC log records |
Detailed Test Cases
Test Case OQ-GLR-001
Procedure: Set the GLR to a predetermined temperature and monitor the system for stability.
Acceptance Criteria: Temperature must remain within ±2°C of the set point for at least 30 minutes.
Evidence: Document temperature logs and any deviations.
Test Case OQ-GLR-002
Procedure: Apply pressure to the GLR and monitor the readings from the pressure gauge.
Acceptance Criteria: Pressure must remain within ±5 psi of the set point.
Evidence: Document pressure logs and any deviations.
Test Case OQ-GLR-003
Procedure: Set the agitation speed and verify the RPM using calibrated equipment.
Acceptance Criteria: RPM must be within ±10% of the set point.
Evidence: Document RPM readings and any deviations.
Test Case OQ-GLR-004
Procedure: Review PLC logs to ensure they accurately reflect the operation of the GLR.
Acceptance Criteria: No discrepancies between PLC logs and manual observations.
Evidence: Document PLC logs and any discrepancies.
Deviations
Any deviations from the acceptance criteria must be documented and investigated. A deviation report must be generated, and corrective actions must be implemented as necessary.
Approvals
This protocol must be reviewed and approved by the Quality Assurance team prior to execution. Signatures below indicate approval of the protocol and its contents.
