Operational Qualification Protocol for Microneedle Mold Filling / Casting System

Objective: To validate the operational performance of the Microneedle Mold Filling / Casting System to ensure it meets specified requirements for producing microneedle arrays.

Scope: This protocol applies to the Microneedle Mold Filling / Casting System used in the R&D and Production areas for the development and manufacturing of transdermal microneedles/advanced patches.

Responsibilities:

  • Validation Team: Execute the OQ protocol and document results.
  • Quality Assurance: Review and approve the OQ protocol and results.
  • Operations: Provide necessary access and support during testing.

Prerequisites:

  • Completion of Installation Qualification (IQ).
  • Availability of all necessary materials and equipment.
  • Training of personnel on the Microneedle Mold Filling / Casting System.

Equipment Description: The Microneedle Mold Filling / Casting System is designed to accurately fill molds with a formulation to create microneedle arrays. It features precision filling capabilities and temperature monitoring to ensure ideal curing conditions.

Test ID Procedure Acceptance Evidence
OQ-001 Verify fill accuracy using calibrated measuring devices. Fill accuracy within ±5% of target volume. Calibration certificates and measurement logs.
OQ-002 Measure curing time at specified temperature. Curing time matches specified range in URS Annex11. Curing time records.
OQ-003 Record temperature during the filling and curing process. Temperature remains within specified limits. Temperature logs and monitoring records.

Detailed Test Cases:

  • Test Case OQ-001: Conduct fill accuracy tests by filling multiple molds and measuring the filled volume. Document the results and calculate the average fill accuracy.
  • Test Case OQ-002: Initiate the curing process and document the time taken for the material to cure at the specified temperature. Compare with acceptance criteria.
  • Test Case OQ-003: Monitor and log the temperature during the filling and curing stages. Ensure that the temperature remains within the established limits throughout the process.

Deviations: Any deviations from the acceptance criteria must be documented and investigated. A corrective action plan should be developed if any critical parameters are not met.
