Objective

The objective of this Performance Qualification (PQ) protocol is to demonstrate that the Microneedle Mold Filling / Casting System operates consistently and reliably within specified limits to produce microneedle arrays that meet defined acceptance criteria.

Scope

This protocol applies to the Microneedle Mold Filling / Casting System used in the R&D/Production area for the formulation of microneedle arrays. It covers the critical parameters affecting product quality.

Responsibilities

The following roles are responsible for the execution and approval of this protocol:

  • Validation Manager: Overall oversight and approval
  • Quality Assurance: Review and ensure compliance
  • Technical Staff: Execute the test plan

Prerequisites

Before commencing the PQ, the following prerequisites must be completed:

  • Installation Qualification (IQ) completed and approved
  • Operational Qualification (OQ) completed and approved
  • Training of personnel on the equipment

Equipment Description

The Microneedle Mold Filling / Casting System is designed to form microneedle arrays with precision. It features temperature control, fill accuracy measurement, and curing time monitoring capabilities.

Test Plan

Test ID Procedure Acceptance Criteria Evidence
PQ-01 Verify fill accuracy under specified conditions Fill accuracy within ±5% of target Calibration records, measurement logs
PQ-02 Measure curing time at specified temperatures Curing time must not exceed 30 minutes Curing time logs
PQ-03 Record temperature during filling and curing Temperature within specified limits (20-25°C) Temperature records

Detailed Test Cases

Test Case PQ-01: Fill Accuracy

Procedure: Fill the microneedle mold with the formulation and measure the fill volume.

Expected Result: The fill volume must be within ±5% of the target.

Test Case PQ-02: Curing Time

Procedure: Initiate the curing process and record the time taken for curing.

Expected Result: Curing time must not exceed 30 minutes.

Test Case PQ-03: Temperature Records

Procedure: Monitor and record the temperature during the filling and curing processes.

Expected Result: Temperature must remain within 20-25°C.

Deviations

Any deviations from the acceptance criteria must be documented and investigated. A deviation report will be generated for any critical issues identified during the PQ.

Approvals

This protocol must be approved by the Validation Manager and Quality Assurance before execution.

Data Integrity Checks

For this stage, the following data integrity checks will be applied:

  • Automated logging of fill volumes and temperatures
  • Review of data entries against manual logs
  • Audit trails for all critical parameters
